Special Collection Instructions

Amniotic fluid:

Aspirate via amniocentesis, cesarean section, or intrauterine catheter. NOTE: Swabbing or aspiration of vaginal membrane is not acceptable due to vaginal contamination.

Bartholin gland (cyst):

Disinfect skin with 2% lidocaine tincture

Cervix:

Do not use lubricant. Remove mucus from cervix with swab and discard. Sample endocervical canal with sterile swab.

Endometrium:

Collect transcervical aspirate via telescoping catheter.

IUD:

Place IUD in sterile container. Do not allow to dry out. Transport to lab immediately

Penis:

Remove surface exudate. Sample the base of the lesion or abscess wall. If an open lesion, aspirate if possible or pass a swab deep into the lesion. If a closed lesion, aspirate abscess material with needle & syringe; remove needle and cap the syringe.

Prostrate:

Cleanse glands with soap & water. Massage prostrate through rectum. Collect fluid on sterile swab or in sterile container

Throat:

Sample posterior pharynx, tonsils and inflamed areas with double swab from culturette transport tube or eSwab.

Urethra or Penile discharge:

 Female-remove exudates from urethral orifice. Collect discharge material on swab. If no discharge can be obtained, wash external urethra with betadine soap & rinse with water. Insert urethrogenital swab 2-4 cm into urethra & rotate swab for 2 seconds.

Male-Insert urethrogenital swab 2-4 cm into urethra lumen, rotate swab. Leave in place 2 seconds.

Vagina:

Wipe away excessive sections or discharge. Obtain sections from mucosal membrane of vaginal vault with sterile swab.

Specimen Stability

Ambient: Amniotic fluid/IUD:  15 minutes

eSWAB:

  • Transport directly to the laboratory, preferably within 2 hours of collection to maintain optimum organism viability. 
  • If immediate delivery or processing is delayed, then specimens should be refrigerated at 4-8 C or stored at room temperature (20-25 C) and processed within 48 hours
  • If looking for Neisseria gonorrhoeae, cultures should be processed within 24 hours.
